Abstract

A mouse monoclonal antibody (Ab 100) to carboxypeptidase A (CPA) which binds to the enzyme with a high affinity constant (˜109 M-1) without affecting its catalytic activity was prepared, purified and characterized. Covalent binding of the monoclonal antibody to Eupergit C, or noncovalent binding to Sepharose-Protein A, yielded carriers reacting specifically with CPA to give immobilized enzyme preparations displaying full catalytic activity and improved stability. The method represents a general technique for the preparation of highly active immobilized enzymes.
