Abstract
The cytotoxic effect of mechlorethamine and cyclophosphamide on the lymphocytes of 21 patients with chronic lymphocytic leukemia was studied by determining the uptake of (methyl-3H)thymidine, (G-3H)uridine and 1-(G-3H) leucine into cell DNA, RNA and protein, respectively, after preincubation with these drugs. DNA synthesis was detected in the lymphocytes of six patients and was inhibited by mechlorethamine in four of them. RNA and protein synthesis were significantly inhibited by this drug (49.4%, 0.001<p<0.05 and 37.5%, 0.02<p<0.025, respectively). With cyclophosphamide only a tendency to inhibition was detected.
