Abstract
A rapid 51Cr release assay was used to study the attacking cell in human antibody dependent cell mediated cytotoxicity. Two kinds of target cells were used: human melanoma tissue culture cells (HML I) and Rh+ human erythrocytes. Serum from multiply transfused patients was the source of antibodies against melanoma cells and serum from patients immunized against Rh+ was used against erythrocytes. Cells separated by Ficoll Hypaque (including 10 to 15% monocytes) effectively attacked antibody treated melanoma cells and erythrocytes. Incubation of attacking cells on a nylon column depleted the ability of mononuclear cells to kill erythrocytes but did not alter lysis of melanoma cells. Granulocytes were not effective in killing cells.
