N2 - Ionic concentration-polarization (CP)-based biomolecule preconcentration is an established method for enhancing the detection sensitivity of target biomolecules immunoassay. However, the formed preconcentrated plug of biomolecules sweeps over the surface immobilized antibodies and results in a short duration of overlap and with this also a smaller binding between target analyte and immobilized molecular probes. To overcome this, we have periodically formed a preconcentrated plug of biomolecules sweeping over the desired region of binding. We found that there is an optimal sweeping condition which results in maximum retention time of the preconcentrated plug over a desired sensing region and enhanced detection sensitivity.
