Abstract

In organ transplantation there remains a relatively high incidence of delayed graft function related with inferior organ preservation and limited storage periods. Captopril (CTP), as an angiotensin converting enzyme (ACE) inhibitor but, unlike other ACE inhibitors, contains sulfhydryl group and can act as free radical scavenger. Therefore, we aimed to investigate the use of Captopril in EuroCaollins and University of Wisconsin preservation solutions. CTP containing (23μM) forms were also used with straight solutions during the perfusion (P) and the storage (S) periods of the kidney cells. The rabbit kidneys were flushed invivo with 100 ml of cold solutions. At the end of the perfusion fine needle aspirations were performed and the cellular aspirates were stored for 72 hours in EC, UW and CTP containing forms. At 24, 48 and 72 hours of the storage we used MTT assay to determine the cellular viability of preserved cells which is an indicator of mitochondrial enzymatic activity. In both solutions, CTP increased the viability of the preserved kidney cells except in use only for storage. The efficacy of EC and UW solutions were upgraded significantly at 24, 48, and 72 hours in groups. CTP showed its distinguished effect when it was used during both perfusion and storage periods. CTP was found very effective to be used in both preservation solutions for prolonging the viability of the kidney cells.
