A rapid and sensitive radioreceptor assay for tertiary amine tricyclic antidepressants

Abstract

Psychotropic drugs including the tricyclic antidepressants are among the most commonly prescribed agents in the world. Recent clinical studies have indicated that careful monitoring of the plasma concentration of the parent drug and their pharmacologically-active metabolites can be useful in adjusting doses for therapeutic purposes as well as for monitoring compliance. Nevertheless, analytical methods for determining the blood level(s) of most antidepressants are cumbersome and not easily applicable to clinical management. With the discovery of high affinity and specific binding sites for radiolabelled tricyclic antidepressants the development of rapid and sensitive methods for determining their tissue levels has become feasible. In this report we describe a radioreceptor assay for measuring the plasma levels of the tricyclic antidepressants and especially their tertiary amine derivatives. This method is rapid and sensitive and could prove useful in the clinical monitoring of these agents.
